Lupron Depot in combination with a progestin, norethindrone acetate 5 mg daily, has been shown to significantly reduce bone loss, a major adverse effect of GnRH agonist therapy. No significant differences in efficacy between Lupron Depot alone and Lupron Depot plus progestin were observed.1,2,3
Reduction of Bone Loss
A study of Lupron Depot plus progestin found that the mean loss of bone density from baseline to 12 months was ≤1%.

The duration of initial treatment or retreatment should be limited to 6 months. All patients in these trials received 1,000 mg/day of calcium supplementation.1
Reduction of Hot Flashes
Additionally, add-back therapy offers the benefit of reducing the number of hot flashes.1
Lupron Depot plus norethindrone acetate 5 mg daily reduces the number of days with hot flashes. The percentage of patients reporting hot flashes decreased from 86% to 58%.1

Lupron Depot plus norethindrone acetate 5 mg daily reduces the number of hot flashes per day. In one study, women taking Lupron Depot plus progestin reported hot flashes on an average of 67% fewer days in month 6 of treatment than those taking Lupron Depot alone.3
